Firstly they would not have been able to come by train;

says here that the transcontinental RR was commisioned in 1862 and completed
in 1869, I was thinking it was later. They could therefore have come to CA
by train and then North, however the fact they are not in the 1860 Census
might be a result of their having been in transit then. More likely they
were simply missed, or are in a part of the census no longer readable or are
indexed by some name that doesn't come up in your searching.

If you'd like I'd be glad to do a search for them, sometimes I get lucky and
if you post their details here someone else might be able to find them also.

As to the death cert, very few places had death certificates prior to the
20th century, ironically Missouri seems to have started the practice fairly
early and they have a lot of stuff online which I assume is where you
looked.

Local newspapers might have some news on her death, or she might be in a
Church record, but not guaranteed. My great great Grandfather Melvin died
sometime around 1831. The only record of his death we have been able to
unearth is a listing of it in the death record for a son. No trace of when
or where he died has ever been turned up.

I have several ancestors who lived in Missour in 1850, can't find in 1860,
then most of them show up in Yamhill County Oregon in 1870.

My question is, would they have travelled by train or by wagon train?  There
were multiple families that came together.  Also, why am I having a hard
time finding them in 1860?  If they were waiting for or in a wagon train,
would they have been missed by the census taker?  I have one ancestor who
lost a wife in Missour approx 1862 and remarried by 1863.  I can't find
death certificate of 1st wife.

Surnames are Lambert and Forrest.
